I spend already quite some time to reproduce and understand the logic of the internal standard normalization using EquiSPLASH and TG 17:0_17:0_17:0 on my raw data, choosing the option "Peak intensity / IS peak". Data Visualization > Normalization > from the drop-down menu Peak intensity / IS peak. From the Alignment result export comparing the output "Raw Data (Area)" and "Normalized Data (Area)" I can see that the normalization has been applied successfully, but the normalized peak areas do not reflect the ratios (division of the peak area by the internal standard peak area of the dedicated lipid class), or am I just not understanding it correctly?
MS-DIAL version is 5.4.241004
Below is an example of peak areas with and without normalization applied. The image below reflects the current settings I am using now. Thanks for any help!
Example MS-DIAL output:
Internal standard (TG 17:0_17:0_17:0) RAW peak area for sample 1: 24745401
TG 46:0 RAW peak area in sample 1: 54470511
